It’s been a while since Twitter implemented something actually useful to it’s service - no, changing the design or adding promoted tweets don’t count as useful stuff -, and this time is Web Notifications, not Desktop Notifications, the difference is that these will only work on Twitter’s website; if you’re browsing another site, you won’t get a notification.
To give you an idea, it’s like Facebook notifications, when you have a new comment or message a little notification will popup to the bottom-right of the page... like that.
To activate, deactivate them, just go to Settings > Web Notifications, let’s just hope to get desktop notifications soon, like TweetDeck.
